Tridem Bioscience has enrolled the first patients in a phase Ib clinical trial of TRB-001, an intradermal therapeutic vaccine targeting misfolded alpha-synuclein in Parkinson’s disease.
The study is evaluating the candidate’s safety and immunogenicity, alongside preliminary biomarkers of efficacy. Top-line results are expected in mid-2027.
TRB-001 is designed to generate an immune response against misfolded alpha-synuclein, which is associated with the pathology of Parkinson’s disease. Current treatments address symptoms, but there are no approved disease-modifying therapies for the condition.
Tridem is also completing analysis of a smaller phase I proof-of-concept study of TRB-001, which has finished recruitment. The study is intended to provide initial data on whether the vaccine can affect its target in the central nervous system. The company expects top-line results in January 2027.
The company also reported observations from its named-patient-use programme, including sustained target-specific antibody levels over more than 80 weeks. Tridem said antibody levels observed in patients were comparable with those achieved in preclinical studies and reached levels it described as being within the range seen with monoclonal antibody therapies.
The company reported a favourable safety profile and said it had observed signs of clinical activity, although the release does not provide efficacy data from the programme.
Tridem is also developing vaccine programmes targeting Alzheimer’s disease, inflammatory skin diseases and atherosclerotic cardiovascular disease (ASCVD), with the company reporting preclinical proof-of-concept data in each area.
Markus Mandler, Chief Executive Officer of Tridem Bioscience, said the start of the phase Ib study marked a significant step for the company and its Parkinson’s programme.
The company said it is seeking co-development and licensing partners for programmes across its pipeline.
